The development of sustainable and efficient synthetic methodologies is a central challenge in modern organic chemistry, particularly for accessing complex molecules under mild and environmentally friendly conditions. Isocyanides represent a uniquely versatile class of C1 building blocks, owing to their distinctive electronic structure and broad reactivity in multicomponent, radical, and insertion reactions. In this context, visible-light-driven processes and solvent-controlled reactivity offer powerful opportunities to expand the synthetic utility of isocyanides. In this thesis, three projects focusing on expanding the sustainable synthetic methodologies have been illustrated to show the versatility of isocyanide as C1 building block.

Chapter 1 presents a general overview of photochemistry from a mechanistic perspective to its application in synthetic chemistry. Besides, isocyanides, as the starting material, an overview of fundamental understanding of isocyanide structure at molecular level and its internation to light and reactivity has been discussed. Isocyanides have been intensively explored in synthetic chemistry since its first discovery by Lieke27 in 1859. Over the past decades, isocyanide chemistry has attracted considerable attention due to its unique reactivity patterns, which have been exploited in drug discovery and material science. Of the various reaction types, multicomponent reactions and transition-metal-catalyzed insertion reactions are the most extensively explored (Scheme 1).

Scheme 1. General summary of isocyanide reaction types
Beside the traditional two electron chemistry via ionic pathways, isocyanides also have shown ability to interact with radicals, forming versatile imidoyl radicals, which are utilized for the synthesis of e.g. functionalized heterocycles, nitriles, imines and amines (Scheme 2).

In Chapter 2, I have disclosed that tryptamine-derived isocyanides could undergo aggregation-induced charge transfer under visible light irradiation, which enables the dearomatization of indoles followed by affording three-dimensional constrained spirocyclic scaffold under mind reaction conditions (Scheme 3).

Tryptamine-derived isocyanides possess an electron-rich indole ring and an electron-deficient isocyanide group. We demonstrated that under visible light, rather than undergoing intramolecular charge transfer, these molecules form intermolecular slip-stacked aggregate complexes. These complexes facilitate a single electron transfer (SET) to generate carbon based-radical intermediates without requiring any external photocatalyst or transition metals. The ability of tryptamine-derived isocyanides to generate radical intermediates through an aggregation-based charge transfer was presented for the first time. Furthermore, we have supported our hypothesis through optical spectroscopy, HRMS and computational studies. Through this methodology we were able to synthesize over 30 spiroindolenines in good to moderate yields with excellent functional group tolerance.

In Chapter 3, I described a catalyst-free, visible-light-driven method for synthesizing thioformimidates from aromatic isocyanides and thiols under mild conditions. building on our previous studies with tryptamine-derived isocyanides from Chapter 2, we discovered that aromatic isocyanides can also undergo aggregation based charge transfer upon visible light irradiation which can further be utilized for the generation of thiyl radicals from aromatic as well as aliphatic thiols, eliminating the need for photocatalysts (Scheme 4). The reaction shows broad substrate scope, high functional-group tolerance, and compatibility with aqueous conditions, providing efficient access to thioformimidates without the common side products associated with earlier methods.

Importantly, the resulting thioformimidates were further used as intermediates for the selective conversion of cysteine-containing peptides into dehydroalanine derivatives, a valuable transformation for peptide and protein modification. Mechanistic studies involving spectroscopy, quenching experiments, and computational analysis suggest that aromatic isocyanide aggregates absorb visible light and initiate radical formation through a proton coupled electron transfer (PCET) process. Overall, the work introduces a sustainable and practical strategy for thioformimidate synthesis, expands the understanding of isocyanide photochemistry, and provides a powerful tool for late-stage peptide functionalization.

In Chapter 4, I demonstrated that using strong hydrogen bond donating (HBD) alcohols, particularly 1,1,1,3,3,3-hexafluoroisopropanol (HFIP), can significantly accelerate the reaction kinetics of the classical Passerini three-component reaction (P-3CR) when used as cosolvent. While conventional alcoholic solvents typically slow down the P-3CR by competing with the carboxylic acid in the rate-determining step, we found that a 20% v/v mixture of HFIP in most aprotic organic solvents, such as dichloromethane and chloroform, can enhance the rate of the reaction, achieving up to a 382-fold increase in chloroform (Scheme 5).

This kinetic acceleration occurs across a broad substrate scope without significantly influencing the overall chemical yields, although the effect is suppressed in strong hydrogen bond accepting (HBA) solvents like DMF. Experimental kinetic studies and DFT calculations provided a mechanistic rationale, indicating that the rate-determining isocyanide addition is optimized when two HFIP molecules participate in hydrogen bonding with the aldehyde, making this approach highly valuable for high-throughput screening applications and sterically hindered or otherwise slow-reacting substrates.

In Chapter 5, future research directions inspired by the work described in this thesis are discussed, together with preliminary results obtained so far. First, based on the mechanistic investigation of tryptamine-derived isocyanides, a general hypothesis is proposed in which charge transfer from the indole ring to tethered isocyanide moiety can happen either inter or intramolecularly. By exploiting this charged-transfer phenomenon, a series of cyclic products could be designed and synthesized selectively. Next, following the successful formation of dehydroalanine (DHA) residues in dipeptides in Chapter 3, preliminary trials with pentapeptides are presented. Based on these results, this methodology is proposed as a potential protection strategy with cysteine in SPPS, addressing the bottlenecks associated with the synthesis of cysteine-containing peptides, e.g. racemization, beta-elimination, and premature oxidation. In addition, inspired by discovery that hydrogen bond donating solvents can accelerate the Passerini reaction when used as cosolvent, the optimized condition are proposed for the synthesis of poly (ester amide)s. Finally, the introduction of disulfide containing side groups into poly (ester amide)s is proposed as a strategy to enhance their biodegradability.
["GET
