Hun gedrag hangt nauw samen met hun stofwisseling: veranderingen in het metabolisme be\u00efnvloeden direct hoe deze cellen functioneren. Aanpassingen in de stofwisseling voorzien niet alleen in de energiebehoefte van macrofagen, maar bepalen ook hun fenotype en hun vermogen om andere cellen te reguleren. Daarom wordt het gericht be\u00efnvloeden van metabolisme gezien als een veelbelovende manier om macrofagen bij ziekte bij te sturen, bijvoorbeeld om de immuunreactie tegen tumoren te versterken. Toch bestaan er nog open vragen over hoe specifieke metabole processen de macrofaagfunctie in het lichaam reguleren, hoe deze processen functioneren in kanker en welke metabole aangrijpingspunten benut kunnen worden.\n\nDit proefschrift onderzoekt deze vragen door fundamenteel en toegepast onderzoek naar macrofaagmetabolisme te combineren. Het werk bestaat uit twee delen: deel 1 richt zich op de fundamentele mechanismen van macrofaagmetabolisme (hoofdstukken 2-5), terwijl deel 2 de rol van macrofaagmetabolisme in kanker onderzoekt (hoofdstukken 6-8).\n\nIn hoofdstuk 2 en 3 wordt beschreven hoe immunometabolieten, stofwisselingsproducten die ophopen bij immuunactivatie, in combinatie met epigenetische processen de functie van macrofagen be\u00efnvloeden. Deze hoofdstukken benadrukken dat metabolieten niet alleen bijproducten van de stofwisseling zijn, maar een actieve rol spelen in het reguleren van macrofaagfunctie.\n\nHoofdstuk 4 beschrijft de ontwikkeling van een praktische experimentele toolbox om het metabolisme van macrofagen systematisch te bestuderen, zowel in muis als in menselijke cellen. Door verschillende metabole en functionele meetmethoden te combineren, helpt deze aanpak onderzoekers om gerichter te kiezen welke technieken het beste passen bij hun onderzoeksvraag.\n\nIn hoofdstuk 5 staat het immunometaboliet D-2-hydroxyglutaraat (D-2HG) centraal. Hier wordt aangetoond dat dit molecuul ophoopt in latere fases van macrofaagactivatie en ontstekingsreacties zowel in kweek- als in diermodellen kan afremmen. De effecten blijken deels onafhankelijk van bekende regulatiemechanismen, wat wijst op aanvullende manieren waarop metabolieten ontsteking kunnen be\u00efnvloeden. Deze resultaten verbreden het spectrum van bekende immunometabolieten en benadrukken dat veranderingen in metabolietniveaus in de tijd een belangrijke rol spelen bij de regulatie van macrofaagfunctie.\n\nHet moduleren van de stofwisseling van macrofagen is bijzonder relevant in de context van kanker, waar macrofagen zich vaak in een metabool uitdagende omgeving bevinden. Deze omstandigheden dragen ertoe bij dat macrofagen in tumoren regelmatig een tumor-ondersteunende rol aannemen in plaats van de kanker te bestrijden.\n\nHoofdstuk 6 bespreekt hoe de tumoromgeving het metabolisme van macrofagen be\u00efnvloedt en hoe metabole interacties tussen kankercellen en macrofagen bijdragen aan tumorontwikkeling.\n\nHoofdstuk 7 onderzoekt het eiwit ATP-citraatlyase (Acly), dat een belangrijke rol speelt in de productie van acetyl-CoA, een bouwsteen voor vetaanmaak en genregulatie. Hoewel het uitschakelen van Acly het gedrag van macrofagen in kweekmodellen verandert, heeft dit in muismodellen geen effect op tumorgroei en leidt het niet tot duidelijke veranderingen in macrofagen in tumoren. Dit benadrukt dat metabole interventies sterk afhankelijk zijn van de biologische context en dat bevindingen uit kweekmodellen niet altijd direct vertaald kunnen worden naar de complexe situatie in vivo.\n\nHoofdstuk 8 verbindt preklinische bevindingen met humane relevantie door single-cell RNA-sequencing datasets van kankerpati\u00ebnten te analyseren. De resultaten laten zien dat macrofagen binnen tumoren metabool zeer divers zijn en dat verschillende subsets eigen metabole programma's met potenti\u00eble therapeutische relevantie vertonen.\n\nGlobaal draagt dit proefschrift bij aan ons begrip van immunometabolisme door inzicht te geven in hoe metabole processen de functie van macrofagen reguleren. Het benadrukt het belang van dynamiek, cellulaire diversiteit en omgevingsfactoren die macrofaagmetabolisme sturen. Door fundamenteel onderzoek en onderzoek gericht op kanker te combineren verdiept dit proefschrift ons begrip van macrofaagmetabolisme en de relevantie ervan voor therapeutische overwegingen rondom macrofaagactivatie en kanker.","summary":"Macrophages are plastic immune cells that play a crucial role in inflammation and disease. Their function is intimately linked to their metabolism: changes in metabolism directly influence how these cells operate. Metabolic adaptations not only meet the energy demands of macrophages but also dictate their phenotype and their ability to regulate other immune cells. Therefore, targeting macrophage metabolism is considered a promising approach to modulate macrophage function in disease, for instance, to enhance anti-tumor immunity. However, open questions remain regarding how specific metabolic processes regulate macrophage function in the body, how these processes operate in cancer, and which metabolic targets can be exploited.\n\nThis thesis investigates these questions by combining fundamental and applied research on macrophage metabolism. The work consists of two parts: Part 1 focuses on the fundamental mechanisms of macrophage metabolism (Chapters 2-5), while Part 2 explores the role of macrophage metabolism in cancer (Chapters 6-8).\n\nChapters 2 and 3 describe how immunometabolites, metabolic products that accumulate upon immune activation, in combination with epigenetic processes, influence macrophage function. These chapters emphasize that metabolites are not merely byproducts of metabolism but play an active role in regulating macrophage function.\n\nChapter 4 describes the development of a practical experimental toolbox to systematically study macrophage metabolism in both mouse and human cells. By combining various metabolic and functional measurement methods, this approach helps researchers to more precisely choose which techniques best suit their research question.\n\nChapter 5 focuses on the immunometabolite D-2-hydroxyglutarate (D-2HG). It demonstrates that this molecule accumulates in later stages of macrophage activation and can inhibit inflammatory responses in both in vitro and in vivo models. Its effects appear partly independent of known regulatory mechanisms, suggesting additional ways in which metabolites can influence inflammation. These results broaden the spectrum of known immunometabolites and emphasize that temporal changes in metabolite levels play an important role in regulating macrophage function.\n\nModulating macrophage metabolism is particularly relevant in the context of cancer, where macrophages often reside in a metabolically challenging environment. These circumstances contribute to macrophages in tumors frequently adopting a tumor-supporting role rather than combating cancer.\n\nChapter 6 reviews the metabolic pressures present in the TME and discusses how these influence TAM phenotype and function. This chapter highlights the complexity of metabolic crosstalk between cancer cells and TAMs, and discusses how these interactions contribute to tumor progression.\n\nChapter 7 experimentally examines ATP-citrate lyase (Acly), a central metabolic enzyme that provides acetyl-CoA for biosynthetic and epigenetic processes, using a myeloid-specific knockout mouse model. While Acly deletion alters IL-4-induced macrophage responses in vitro, it does not affect tumor growth or overtly reprogram TAM phenotypes in vivo. These findings emphasize the strong context dependence of metabolic interventions and illustrate the limitations of translating in vitro polarization models directly to the tumor setting.\n\nChapter 8 bridges preclinical findings to human relevance by analyzing single-cell RNA sequencing datasets from cancer patients. This analysis reveals substantial metabolic heterogeneity among distinct TAM subsets and identifies subset-specific metabolic programs with potential therapeutic relevance.\n\nTogether, this thesis contributes to the field of immunometabolism by providing mechanistic insight into how metabolic pathways regulate macrophage function in distinct biological contexts. It highlights the importance of temporal dynamics, cellular heterogeneity, and environmental cues in shaping macrophage metabolism. By integrating fundamental and cancer-focused studies, this work refines our understanding of macrophage metabolism and its relevance for therapeutic considerations in macrophage activation and cancer.","auteur":"Kyra de Goede","auteur_slug":"kyra-de-goede","publicatiedatum":"7 december 2026","taal":"EN","url_flipbook":"https:\/\/ebook.proefschriftmaken.nl\/ebook\/kyradegoede?iframe=true","url_download_pdf":"https:\/\/ebook.proefschriftmaken.nl\/download\/5b1c8d3b-e881-4ccd-8f8d-a5be0d6a1bb3\/highres","url_epub":"","ordernummer":"19799","isbn":"978-94-6534-732-5","doi_nummer":"10.5463\/thesis.1890","naam_universiteit":"Vrije Universiteit Amsterdam","afbeeldingen":28520,"video_url":"","podcast_url":"","naam_student:":"","binnenwerk":"","universiteit":"Vrije Universiteit Amsterdam","cover":"","afwerking":"","cover_afwerking":"","design":""},"_links":{"self":[{"href":"https:\/\/www.proefschriftmaken.nl\/en\/wp-json\/wp\/v2\/us_portfolio\/28519","targetHints":{"allow":["GET"]}}],"collection":[{"href":"https:\/\/www.proefschriftmaken.nl\/en\/wp-json\/wp\/v2\/us_portfolio"}],"about":[{"href":"https:\/\/www.proefschriftmaken.nl\/en\/wp-json\/wp\/v2\/types\/us_portfolio"}],"wp:featuredmedia":[{"embeddable":true,"href":"https:\/\/www.proefschriftmaken.nl\/en\/wp-json\/wp\/v2\/media\/28520"}],"wp:attachment":[{"href":"https:\/\/www.proefschriftmaken.nl\/en\/wp-json\/wp\/v2\/media?parent=28519"}],"wp:term":[{"taxonomy":"university_us_portfolio","embeddable":true,"href":"https:\/\/www.proefschriftmaken.nl\/en\/wp-json\/wp\/v2\/university_us_portfolio?post=28519"}],"curies":[{"name":"wp","href":"https:\/\/api.w.org\/{rel}","templated":true}]}}
