Zo\u2019n overgang verandert de selectiedruk op eigenschappen die betrokken zijn bij voortplanting. Seksuele kenmerken die niet langer nodig zijn, kunnen daardoor geleidelijk achteruitgaan en uiteindelijk verdwijnen. Wanneer aseksuele voortplanting wordt veroorzaakt door een endosymbiotisch organisme (een organisme dat in of binnen de cellen van een ander organisme leeft), kan dit organisme functies van de gastheer overnemen of compenseren die door de overgang naar aseksualiteit overbodig zijn geworden. Dit verschijnsel staat bekend als gecompenseerd eigenschapsverlies.\n\nDit proefschrift onderzoekt de gevolgen van de overgang naar aseksuele voortplanting, met bijzondere aandacht voor gecompenseerd eigenschapsverlies. Als modelsysteem is de springstaart Folsomia candida gebruikt, samen met diens endosymbiont Wolbachia. Wolbachia is een bacterie die bekendstaat om het be\u00efnvloeden van de voortplanting van zijn gastheren. Er zijn sterke aanwijzingen dat deze bacterie ook de aseksuele voortplanting van F. candida veroorzaakt. In dit proefschrift wordt de overgang naar parthenogenese (een specifieke vorm van aseksuele voortplanting vanuit onbevruchte eicellen) onderzocht vanuit drie invalshoeken: het fenotype, de symbiose en het genoom. Samen geven deze perspectieven inzicht in de processen achter deze reproductieve omslag en in de gevolgen ervan voor de genomen van zowel gastheer als symbiont.\n\nHet onderzoek begon met de vraag wat er gebeurt met mannelijke kenmerken die in parthenogenetische lijnen niet langer functioneel of noodzakelijk zijn. Een vergelijking van seksuele en aseksuele populaties van F. candida liet zien dat verschillende belangrijke reproductieve eigenschappen in aseksuele lijnen sterk zijn achteruitgegaan. Dit gold met name voor de hoeveelheid sperma in de spermatofoor (bolletje met sperma dat kan worden opgepakt door het vrouwtje om haar eitjes te bevruchten) en voor de aantrekkingskracht van spermatoforen op basis van geurstoffen. De mate en het patroon van deze achteruitgang verschilden tussen lijnen. Dit past bij de theorie van neutrale mutatieaccumulatie, waarbij zodra de natuurlijke selectie op een eigenschap niet langer in stand wordt gehouden willekeurige mutaties kunnen ophopen en uiteindelijk functieverlies optreedt. Wanneer Wolbachia inderdaad de parthenogenetische voortplanting mogelijk maakt, kan het verdwijnen van deze mannelijke eigenschappen worden gezien als gecompenseerd eigenschapsverlies.\n\nVervolgens richtte het onderzoek zich op de endosymbiont die waarschijnlijk verantwoordelijk is voor deze reproductieve omslag: de Wolbachia-stam wFol. De overgang naar aseksualiteit wordt vaak gekoppeld aan een stabiele, langdurige endosymbiotische infectie. Experimenten met transinfectie en hittestress lieten echter zien dat de relatie tussen gastheer en symbiont complexer is. Hoge temperaturen verlaagden weliswaar het aantal Wolbachia-bacteri\u00ebn, maar activeerden niet de lytische cyclus van de bijbehorende bacteriofaag WO. Daarnaast bleek het moeilijk om stabiele Wolbachia-infecties tot stand te brengen in seksuele lijnen van F. candida. Dit wijst erop dat de aangeboren afweer van de gastheer een belangrijke belemmering kan vormen voor horizontale overdracht van de symbiont. Daardoor kan dit mechanisme ook bijdragen aan het in stand houden van de evolutionaire scheiding tussen seksuele en aseksuele populaties.\n\nOp genomisch niveau laat dit proefschrift zien dat de evolutie van endosymbionten niet altijd verloopt volgens het klassieke patroon van genoomreductie. Bij obligate symbiose wordt vaak verwacht dat een symbiontgenoom kleiner en eenvoudiger wordt doordat overbodige genen verloren gaan. Het genoom van wFol bleek echter uitzonderlijk groot. Dit genoom bevat sterke uitbreidingen van regio\u2019s die zijn afgeleid van bacteriofagen. Ook is het verrijkt met genen die betrokken zijn bij DNA-herstel. Deze kenmerken wijzen eerder op het behoud van genetisch materiaal dan op vergaande genoomreductie.\n\nEen vergelijkbaar patroon is zichtbaar in de gastheer. Seksuele populaties van F. candida behouden een relatief gestroomlijnde genoomarchitectuur, terwijl aseksuele lijnen juist een aanzienlijke uitbreiding van hun genoom vertonen. Die uitbreiding gaat samen met een grotere ophoping en retentie van repetitieve DNA-elementen, duplicatie van genoomsegmenten en resten van mitochondriaal DNA. Zowel in de symbiont als in de aseksuele gastheer lijkt genoombehoud en -uitbreiding dus een belangrijker rol te spelen dan genoomstroomlijning.\n\nSamengevat laat dit proefschrift zien dat de overgang naar parthenogenese in F. candida niet alleen leidt tot het verlies van seksuele eigenschappen. De overgang gaat ook gepaard met ingrijpende veranderingen in de relatie tussen gastheer en symbiont en met een grootschalige herstructurering van beide genomen. In aseksuele lijnen lijkt het verlies van seksuele functies te worden opgevangen door Wolbachia en door een genoomarchitectuur die de accumulatie en het behoud van genetisch materiaal bevordert. Deze verschuiving van stroomlijnen naar behoud en uitbreiding kan mogelijk helpen om de gevolgen van het ontbreken van seksuele recombinatie op te vangen. Daarmee levert dit onderzoek nieuwe inzichten op in de evolutie van genomen tijdens grote veranderingen in voortplantingswijze en levenscyclus.","summary":"The evolutionary transition from sexual to asexual reproduction represents one of the most profound lifestyle shifts across the tree of life, causing large shifts in selective pressures on phenotypic and genotypic traits. These shifts in pressures could lead to the decay and disappearance of sexual traits no longer needed for asexual reproduction. When asexual reproduction is induced by a symbiotic organism, that organism may compensate for host functions that become redundant after the loss of sexual reproduction. This process, in which the symbiont replaces or maintains functions formerly supported by sexual traits or processes, is termed compensated trait loss. This thesis investigates the multifaceted consequences of the transition to asexuality, with particular emphasis on compensated trait loss, using the springtail Folsomia candida and its endosymbiont Wolbachia as a study system. Wolbachia is known for its reproductive manipulations and there are strong indications it is involved in the asexual reproduction of F. candida. Its transition to reproduction through parthenogenesis was examined through phenotypic, symbiotic, and genomic lenses, to get a full scope of the involved processes and genomic consequences.\n\nThe investigation began at the phenotypic level, focusing on the fate of male-specific traits that become redundant in parthenogenetic lineages. Through a comparative analysis of sexual and asexual F. candida populations, it was demonstrated that several key reproductive traits, specifically sperm DNA quantity and the olfactory attractiveness of spermatophores, underwent significant degradation in asexual lines. The observed patterns of decay, which varied between lineages, are highly consistent with the neutral mutation accumulation theory, suggesting that once the selective pressure to maintain these traits is removed, stochastic loss-of-function mutations become the primary driver of phenotypic deterioration. It can be argued that this is a form of compensated trait loss, given the assumption that Wolbachia is allowing parthenogenetic reproduction, which would compensate for these phenotypic traits that are now lost.\n\nBuilding upon this understanding of trait decay, the research then turned to the endosymbiont driving this reproductive shift: the Wolbachia strain wFol. While the transition to asexuality is often linked to the stability of an endosymbiotic infection, our experiments with transinfection and heat stress revealed a complex interplay between host immunity and symbiont persistence. Notably, we observed that while high temperatures directly reduced Wolbachia titres, they did not necessarily trigger the lytic cycle of the associated bacteriophage WO. Furthermore, the difficulty in establishing stable infections in sexual lineages suggests that the host\u2019s innate immune response may act as a significant barrier to horizontal symbiont transfer, potentially reinforcing the evolutionary boundaries between sexual and asexual populations.\n\nSubsequently, the exploration proceeded at the genomic level where it challenged established paradigms regarding endosymbiont evolution. Contrary to the long-standing symbiont reduction model, which predicts genome streamlining in obligate relationships, we discovered that the wFol genome is exceptionally large and characterized by massive expansions of phage-derived regions. This unexpected expansion, accompanied by an enrichment of DNA-repair and ankyrin-repeat genes, suggests a mode of genome preservation rather than reduction. This pattern of genomic stability in the endosymbiont was mirrored in the host; our comparative analysis of F. candida lineages revealed that while sexual populations maintain a streamlined architecture, asexual lineages exhibit significant genomic expansion. This is characterized by an increased accumulation and retention of repetitive elements, segmental duplications, and mitochondrial DNA remnants.\n\nUltimately, this thesis hypothesises that the transition to parthenogenesis in F. candida is not merely a process of trait loss, but one of complex genomic restructuring. The decay of sexual traits is compensated for by a unique evolutionary strategy in asexual lineages: the adoption of a genome architecture optimized for the accumulation and preservation of genetic material. This shift from purging to conservation likely serves as an adaptive mechanism to navigate the lack of sexual recombination, providing a window into the broader dynamics of genome evolution during major life-history transitions.","auteur":"Dr\u00e9 Kampfraath","auteur_slug":"dr-kampfraath","publicatiedatum":"25 november 2026","taal":"EN","url_flipbook":"https:\/\/ebook.proefschriftmaken.nl\/ebook\/drkampfraath?iframe=true","url_download_pdf":"https:\/\/ebook.proefschriftmaken.nl\/download\/a468159b-0860-4d2e-8065-9f46b8bffc91\/highres","url_epub":"","ordernummer":"19767","isbn":"978-94-6534-728-8","doi_nummer":"10.5463\/thesis.1972","naam_universiteit":"Vrije Universiteit Amsterdam","afbeeldingen":28467,"video_url":"","podcast_url":"","naam_student:":"","binnenwerk":"","universiteit":"Vrije Universiteit Amsterdam","cover":"","afwerking":"","cover_afwerking":"","design":""},"_links":{"self":[{"href":"https:\/\/www.proefschriftmaken.nl\/en\/wp-json\/wp\/v2\/us_portfolio\/28003","targetHints":{"allow":["GET"]}}],"collection":[{"href":"https:\/\/www.proefschriftmaken.nl\/en\/wp-json\/wp\/v2\/us_portfolio"}],"about":[{"href":"https:\/\/www.proefschriftmaken.nl\/en\/wp-json\/wp\/v2\/types\/us_portfolio"}],"wp:featuredmedia":[{"embeddable":true,"href":"https:\/\/www.proefschriftmaken.nl\/en\/wp-json\/wp\/v2\/media\/28467"}],"wp:attachment":[{"href":"https:\/\/www.proefschriftmaken.nl\/en\/wp-json\/wp\/v2\/media?parent=28003"}],"wp:term":[{"taxonomy":"university_us_portfolio","embeddable":true,"href":"https:\/\/www.proefschriftmaken.nl\/en\/wp-json\/wp\/v2\/university_us_portfolio?post=28003"}],"curies":[{"name":"wp","href":"https:\/\/api.w.org\/{rel}","templated":true}]}}
