Lichtgebaseerde benaderingen zijn naar voren gekomen om deze uitdaging aan te gaan, waardoor dynamische controle van biologische doelen mogelijk wordt terwijl bijwerkingen worden verminderd. Onder deze benaderingen maakt fotofarmacologie gebruik van lichtgevoelige liganden waarvan de biologische activiteit reversibel kan worden gecontroleerd door bestraling met licht. Fotoschakelbare liganden bieden de mogelijkheid om de eiwitfunctie met hoge precisie te moduleren, wat ze aantrekkelijke verbindingen maakt voor onderzoek. Ondanks aanzienlijke vooruitgang in het veld, vertrouwen veel fotoschakelbare liganden nog steeds op de azobenzeen-structuur, en het omschakelen van deze groep van de trans- naar de cis-configuratie vereist over het algemeen ultraviolette (UV) bestraling. Als gevolg hiervan is hun toepasbaarheid in levende systemen beperkt, aangezien UV-golflengten worden geabsorbeerd voordat ze diepere weefsellagen kunnen bereiken.\n\nG-eiwitgekoppelde receptoren (GPCR's) vormen een van de belangrijkste klassen van geneesmiddeldoelen en zijn een belangrijk aandachtspunt geworden van fotofarmacologisch onderzoek. Verschillende fotoschakelbare liganden voor histaminereceptoren zijn al in het verleden ontwikkeld. Voortbouwend op dit fundament verkent dit proefschrift het ontwerp, de synthese en de karakterisering van de volgende generatie fotoschakelbare GPCR-liganden. Bijzondere nadruk wordt gelegd op het uitbreiden van de fotofarmacologische gereedschapskist buiten de conventionele azobenzenen door alternatieve heteroaryl-fotoschakelaars te onderzoeken en te evalueren hoe deze structurele modificaties zowel fotochemische als farmacologische eigenschappen be\u00efnvloeden. Samen beogen deze studies bij te dragen aan de ontwikkeling van effectievere en biologisch compatibele fotoschakelbare liganden voor GPCR-fotofarmacologie.\n\nHoofdstuk 2 beschrijft de ontwikkeling van een tweede generatie reeks van op arylazopyrazool gebaseerde histamine H3-receptor (H3R) antagonisten, ontworpen om eigenschappen te verbeteren in vergelijking met de eerdere fotoschakelbare azobenzeen-liganden. Het introduceren van de arylazopyrazool-fotoschakelaar leidde tot effici\u00ebntere fotoschakeling, betere oplosbaarheid en verbeterde binding aan H3R dan de op azobenzeen gebaseerde template-ligand. Het optimalisatieproces leidde tot de identificatie van de belangrijkste verbinding VUF26063, die picomolaire H3R-binding in zijn trans-isomeer combineert met een uitgesproken afname van affiniteit bij conversie naar de cis-toestand. Een belangrijk structureel kenmerk van dit ligand maakte het ook mogelijk om een tritium-label te introduceren, waardoor het eerste radioactief gelabelde fotoschakelbare ligand werd gegenereerd. Dit instrument werd gebruikt om de kinetiek van dit ligand te bestuderen, wat aangeeft dat het fotochemisch isomeriseert terwijl het zich in de receptor-pocket bevindt.\n\nHoofdstuk 3 introduceert 'photoswitch hop' als ontwerpstrategie. De fenylring van een op azobenzeen gebaseerde H3R-antagonist wordt gesubstitueerd met een reeks heteroaryl-azo (HAA) groepen om te onderzoeken hoe de incorporatie van heteroaromatische ringen de fotochemische, fysicochemische en farmacologische eigenschappen van de resulterende liganden be\u00efnvloedt. De studie onthulde dat het uitwisselen van de fotoschakelaar-structuur de eigenschappen diepgaand kan be\u00efnvloeden, wat resulteert in verbindingen met roodverschoven schakelgolflengten, verbeterde fotostationaire toestanden en een breed scala aan thermische relaxatietijden. Hoewel geen enkele HAA-fotoschakelaar beter presteert dan azobenzeen op alle parameters, vertonen verschillende analogen een superieure receptoraffiniteit en verbeterde schakelgolflengten. In het bijzonder vertoont het arylazoisoxazool-ligand VUF26257 een picomolaire H3R-affiniteit samen met een 13-voudige lichtge\u00efnduceerde affiniteitsverschuiving.\n\nHoofdstuk 4 richt zich op de histamine H1-receptor (H1R), een klinisch belangrijke GPCR waarvoor nog geen voldoende effectief fotoschakelbaar ligand is ontwikkeld. Desloratadine werd gebruikt als template-scaffold voor een 'azoscan'-benadering, waarbij azobenzeen-eenheden werden ge\u00efnstalleerd op beschikbare posities, wat negen liganden opleverde. Onder deze bleek het op de 2-positie gesubstitueerde analoog de meest veelbelovende kandidaat.\n\nHoofdstuk 5 onderzoekt hoe de positie en ori\u00ebntatie van een arylazopyrazool-fotoschakelaar de fotochemische en farmacologische eigenschappen van een ligand be\u00efnvloedt. Ondanks de uitstekende fotochemische eigenschappen, vertoonden de drie bereide verbindingen geen affiniteit voor de H3R, waarschijnlijk door de sterische hinder van de arylazopyrazoolring.\n\nHoofdstuk 6 bespreekt de algemene bevindingen. In totaal verkende dit proefschrift de synthese en karakterisering van de volgende generatie fotoschakelbare liganden voor histaminereceptoren, met nadruk op scaffolds buiten azobenzeen. De bevindingen bevorderen het begrip van hoe lichtgevoelige liganden kunnen worden ontworpen voor ruimtelijke en temporele controle van GPCR's.","summary":"The ability to control biological processes with high spatial and temporal precision remains a major challenge in modern drug discovery. Light-based approaches have emerged to address this challenge, enabling dynamic control of biological targets while reducing off-target effects. Among these approaches, photopharmacology uses light-responsive ligands the biological activity of which can be reversibly controlled through irradiation with light. Photoswitchable ligands offer the possibility to modulate protein function with high precision, making them attractive compounds for research. Despite substantial advances in the field, many photoswitchable ligands still rely on the azobenzene scaffold, and switching this group from its trans to its cis configuration generally calls for ultraviolet (UV) irradiation. As a result, their applicability in living systems is limited, since UV wavelengths are absorbed before they can reach deeper tissue layers.\n\nG protein-coupled receptors (GPCRs) constitute one of the most important classes of drug targets and have become a major focus of photopharmacological research. Several photoswitchable ligands for histamine receptors have already been developed in the past. Building on this foundation, this thesis explores the design, synthesis, and characterization of next-generation photoswitchable GPCR ligands. Particular emphasis is placed on expanding the photopharmacological toolbox beyond conventional azobenzenes by exploring alternative heteroaryl photoswitches and evaluating how these structural modifications influence both photochemical and pharmacological properties. Together, these studies aim to contribute to the development of more effective and biologically compatible photoswitchable ligands for GPCR photopharmacology.\n\nChapter 2 describes the development of a second-generation series of arylazopyrazole-based histamine H3 receptor (H3R) antagonists, designed to improve properties compared to the earlier azobenzene photoswitchable ligands. Introducing the arylazopyrazole photoswitch led to more efficient photoswitching, better solubility, and improved binding to H3R than the azobenzene-based template ligand. The optimization process led to the identification of key compound VUF26063, which combines picomolar H3R binding in its trans isomer with a pronounced erosion of affinity upon conversion to the cis state. A key structural feature of this ligand also made it possible to introduce a tritium label, generating the first radiolabelled photoswitchable ligand. This tool was used to study the kinetics of this ligand, indicating that it photochemically isomerizes while residing inside the receptor pocket.\n\nChapter 3 introduces a 'photoswitch hop' as a design strategy. The phenyl ring of an azobenzene-based H3R antagonist is substituted with a range of heteroaryl azo (HAA) groups to examine how the incorporation of heteroaromatic rings affects the photochemical, physicochemical and pharmacological properties of the resulting ligands. The study revealed that exchanging the photoswitch scaffold can profoundly influence properties, yielding compounds with red-shifted switching wavelengths, improved photostationary states, and a wide range of thermal relaxation times.\n\nChapter 4 targets the histamine H1 receptor (H1R), a clinically important GPCR for which no sufficiently effective photoswitchable ligand has yet been developed. Desloratadine was used as the template scaffold for an \"azoscan\" approach. Among these, the 2-substituted analogue emerged as the most promising candidate.\n\nChapter 5 examines how the position and orientation of an arylazopyrazole photoswitch affects the photochemical and pharmacological properties of a ligand.
