Pogingen om aan deze groeiende behoeften te voldoen, hebben geleid tot verschillende mondiale uitdagingen, waaronder de uitputting van niet-hernieuwbare hulpbronnen, klimaatverandering en milieuvervuiling. Binnen het opkomende concept van een Biobased Economy worden microalgen erkend als een veelbelovende hernieuwbare biomassabron waaruit een breed scala aan voedingsmiddelen en andere producten kan worden afgeleid, zoals cosmetica, supplementen, diervoeder, chemicali\u00ebn, meststoffen en biobrandstoffen. De kweek van microalgen biedt meerdere voordelen, zoals productie gedurende het hele jaar, hoge groeisnelheden, hoge eiwit- en olieopbrengsten, en de mogelijkheid om brak- of zeewater, niet-bouwland of offshore-locaties en teruggewonnen nutri\u00ebnten en CO2 uit afvalstromen te gebruiken. Niettemin moeten de productiekosten van microalgen verder worden verlaagd voor grootschalige toepassingen, met name voor bulkproducten. De economische haalbaarheid kan worden verbeterd door een combinatie van inspanningen om de biomassaproductiviteit te maximaliseren, de input te minimaliseren en alle componenten van de biomassa te valoriseren.\n\nHet onderzoeksdoel van dit proefschrift was het onderzoeken van een nieuw teeltsysteem voor microalgen op locaties op lage breedtegraden, waar overvloedig zonlicht en een constant klimaat gunstige omstandigheden bieden voor de productie van microalgenbiomassa gedurende het hele jaar. De focus van het onderzoek lag op het verbeteren van de biomassaopbrengst op licht en de biomassaproductiviteit per oppervlakte, en op het elimineren van de noodzaak voor actieve temperatuurregeling, wat beide kan helpen de totale biomassaproductiekosten te verlagen.\n\nIn Hoofdstuk 2 wordt een nieuw V-vormig fotobioreactorontwerp voorgesteld, bestaande uit twee reactorpanelen die naar het noorden en zuiden zijn geneigd, om zonlicht op lage breedtegraden op te vangen en te verdunnen. Er werd een wiskundig model ontwikkeld om de inval van zonlicht in een V-vormige fotobioreactor gedurende verschillende dagen van het jaar te simuleren, waaruit de maximale biomassaproductiviteit werd geschat voor Chlorella sorokiniana op Bonaire, uitgaande van onbewolkte omstandigheden en licht-gelimiteerde groei. De modelresultaten laten zien dat biomassaproductiviteiten van 38,3-50,5 g m-2 d-1 en fotosynthetische effici\u00ebnties van 2,5-3,3% kunnen worden bereikt in V-vormige fotobioreactoren, waarbij de waarden toenemen met de hellingshoeken van de reactorpanelen. De biomassaproductiviteiten in V-vormige fotobioreactoren worden geschat op 1,4 keer hoger dan die in een vlakke horizontale fotobioreactor, voornamelijk dankzij lichtverdunning als gevolg van de V-vormige configuratie. De intensiteit van het zonlicht op het middaguur bleek te worden verdund van waarden rond 2000 \u03bcmol PAR-fotonen m-2 s-1 naar gemiddelde waarden van bijna 500 \u03bcmol PAR-fotonen m-2 s-1, afhankelijk van de dag van het jaar en de hellingshoeken van de reactorpanelen. De modelresultaten tonen het potentieel aan van het V-vormige fotobioreactorontwerp voor verbeterde lichtverdunning en biomassaproductiviteit per oppervlakte.\n\nIn Hoofdstuk 4 worden buitenexperimenten beschreven waarbij een prototype van een V-vormige fotobioreactor werd getest voor de kweek van Picochlorum sp. (BPE23) op Bonaire. Picochlorum sp. (BPE23) is een lokale thermotolerante microalg die werd ge\u00efsoleerd uit een zoutwaterlichaam op Bonaire en geselecteerd was op zijn hoge groeisnelheid bij 40\u00b0C. De biomassaproductiviteit van de microalgen werd bepaald bij verschillende verdunningsgraden in continue verdunnings- en herhaalde batch-experimenten, zonder enige vorm van temperatuurregeling. Terwijl de temperaturen overdag opliepen tot 35-45 \u00b0C, werden opmerkelijke gemiddelde biomassaproductiviteiten van 28-31 g m-2 d-1 en fotosynthetische effici\u00ebnties van 3,5-4,3% bereikt in de continue verdunningsexperimenten. In de herhaalde batch-experimenten ervoeren de microalgen waarschijnlijk stress als gevolg van plotselinge veranderingen in licht en temperatuur na dagelijkse verdunning. Er werden zuurstofproductiemetingen uitgevoerd bij verschillende temperaturen om de maximale fotosynthesesnelheden van de reactorculturen te bepalen. Dichte culturen van Picochlorum sp. (BPE23) werden gekenmerkt door hoge maximale fotosynthesesnelheden in vergelijking met eerder gerapporteerde gegevens voor Picochlorum en snelheden van andere microalgen, met name bij een temperatuur van 39\u00b0C. In andere zuurstofevolutie-experimenten en in het laboratorium uitgevoerde groeiexperimenten in vlakke fotobioreactoren bleek Picochlorum sp. (BPE23) ook gekenmerkt te worden door een hoge specifieke groeisnelheid en een laag onderhoudsverbruik vergeleken met andere microalgensoorten, zoals Neochloris oleoabundans en Nannochloropsis sp. (Hoofdstuk 3). De resultaten in Hoofdstuk 3 en 5 benadrukken de robuustheid en het potentieel van Picochlorum sp. (BPE23) voor snelle groei onder wisselende buitenomstandigheden.\n\nIn Hoofdstuk 5 wordt de mogelijkheid van passieve temperatuurregeling in een nieuwe V-vormige fotobioreactor onderzocht voor een locatie op een lage breedtegraad. Er werd een wiskundig temperatuurmodel ontwikkeld om de reactortemperatuur en de relevante warmtefluxen gedurende het jaar op Bonaire te simuleren. Er werden twee scenario's gesimuleerd: \u00e9\u00e9n waarin de V-vormige fotobioreactor op het land wordt gebruikt, en \u00e9\u00e9n waarin de fotobioreactor drijft op water in een bassin. De gemiddelde temperatuur van de landgebaseerde fotobioreactor bleek 35\u00b0C te zijn, terwijl de gemiddelde temperatuur van de drijvende fotobioreactor 32\u00b0C was. Desalniettemin was de temperatuurfluctuatie in een drijvende fotobioreactor (i.e. 30-33\u00b0C) veel kleiner dan die in de landgebaseerde reactor (i.e. 30-42\u00b0C) vanwege het bufferende effect van de watermassa. In een worstcasescenario, waarbij wordt aangenomen dat de convectie boven en onder de reactor beperkt is, bleef de temperatuur in een drijvende fotobioreactor grotendeels onder de 40\u00b0C, terwijl de temperatuur in de landgebaseerde reactor opliep tot 48\u00b0C. Toch bleken de geschatte reactortemperaturen voor zowel de landgebaseerde als de drijvende fotobioreactor geschikt voor de kweek van mesofiele en temperatuurtolerante microalgen zoals Picochlorum sp. zonder temperatuurregeling.\n\nIn Hoofdstuk 6 worden de belangrijkste bevindingen van dit proefschrift gepresenteerd en besproken. Daarnaast wordt een schatting gemaakt van de maximale theoretische biomassaproductiviteit van Picochlorum sp., Neochloris oleoabundans en Nannochloropsis sp. in een grootschalige V-vormige fotobioreactor op Bonaire. Deze schatting was gebaseerd op de biologische parameters bepaald in Hoofdstuk 3 en het wiskundige model ontwikkeld in Hoofdstuk 2. Deze productiviteiten bleken respectievelijk 46,5, 49 en 39 g m-2 d-1 te zijn, wat het hoge groeipotentieel van de Bonairiaanse Picochlorum-stam aantoont. De resultaten benadrukken het belang van kennis over de lichtintensiteit en temperatuur waaraan microalgenculturen worden blootgesteld in een specifiek teeltsysteem en op een specifieke locatie gedurende de dag en het jaar. Op basis van deze kennis moeten microalgenstammen worden geselecteerd die optimaal presteren onder die omstandigheden om de biomassaproductiviteit te maximaliseren.\n\nIn een laatste stap wordt de nieuwe V-vormige fotobioreactor in Hoofdstuk 6 ge\u00ebvalueerd met behulp van een techno-economisch model en een SWOT-analyse. De biomassaproductiekosten voor Picochlorum sp. (BPE23) in een V-vormige fotobioreactor op Bonaire werden geschat op 2,24 \u20ac kg-1 drooggewicht bij een productieschaal van 100 ha en 7,05 \u20ac kg-1 drooggewicht bij een schaal van 1 ha. Deze kosten waren lager dan de biomassaproductiekosten in andere systemen op dezelfde locatie, dankzij de relatief hoge fotosynthetische effici\u00ebntie die kan worden bereikt in een V-vormige fotobioreactor en de afwezigheid van actieve temperatuurregeling. Om de biomassaproductiekosten nog verder te verlagen en de commercialisering van microalgenbiomassa te bevorderen, moeten verschillende mogelijkheden verder worden onderzocht, zoals het gebruik van hernieuwbare energie (bijv. zon- en\/of windenergie), het gebruik van directe luchtafvang of rookgas als bron van CO2, en de ontwikkeling van bioraffinaderij-benaderingen en producten of ervaringen met een hogere toegevoegde waarde. Dit hoofdstuk sluit af met de belangrijkste conclusies en aanbevelingen van dit proefschrift.","summary":"As the Earth\u2019s population continues to grow, worldwide demands for food, fresh water, energy and materials rapidly increase. Attempts to meet these growing demands have led to several global challenges, including the depletion of non-renewable resources, climate change, and environmental pollution. Within the emerging concept of a Biobased Economy, microalgae have been recognized as a promising renewable biomass source from which a wide range of food and non-food products can be derived, such as cosmetics, supplements, feed, chemicals, fertilizers, and biofuels. The cultivation of microalgae can offer multiple advantages, such as year-round production, high growth rates, high protein and oil yields, and the possibility to use brackish or seawater, non-arable land or offshore locations, and reclaimed nutrients and CO2 from waste streams. Nevertheless, the production costs of microalgae need to be further reduced for large-scale applications, particularly for bulk products. Economic feasibility can be improved through a combination of efforts to maximize biomass productivity, minimize inputs, and valorize all components of the biomass.\n\nThe research aim of this thesis has been to investigate a novel cultivation system for microalgae at low-latitude locations, where abundant sunlight and constant climate facilitate favorable conditions for year-round production of microalgal biomass. The focus of the research has been to improve biomass yield on light and areal biomass productivity, and to eliminate the need for active temperature control, both of which can both help lower overall biomass production costs.\n\nIn Chapter 2, a novel V-shaped photobioreactor design is proposed, consisting of two reactor panels inclined towards the north and south, to capture and dilute sunlight at low latitudes. A mathematical model was developed to simulate sunlight entering a V-shaped photobioreactor during several days of the year, from which the maximal biomass productivity was estimated for Chlorella sorokiniana on Bonaire, assuming clear-sky conditions and light-limited growth. The model results show that biomass productivities of 38.3-50.5 g m-2 d-1 and photosynthetic efficiencies of 2.5-3.3% can be achieved in V-shaped photobioreactors, with values increasing with the inclination angles of the reactor panels. The biomass productivities in V-shaped photobioreactors are estimated to be 1.4 times higher than those in a flat horizontal photobioreactor, primarily due to light dilution resulting from the V-shaped configuration. The intensity of sunlight at midday was found to be diluted from values of around 2000 \u03bcmol PAR photons m-2 s-1 to average values down to nearly 500 \u03bcmol PAR photons m-2 s-1, depending on the day of the year and the inclination angles of the reactor panels. The model results show the potential of the V-shaped photobioreactor design for improved light dilution and areal biomass productivity.\n\nIn Chapter 4, outdoor experiments are described in which a prototype of a V-shaped photobioreactor was tested for the cultivation of Picochlorum sp. (BPE23) on Bonaire. Picochlorum sp. (BPE23) is a local thermo-tolerant microalgae that was isolated from a saltwater body on Bonaire and had been selected for its high growth rate at 40\u00b0C. The biomass productivity of the microalgae was determined at different dilution rates in outdoor continuous dilution and repeated batch experiments, without any form of temperature control. While temperatures increased to 35-45 \u00b0C during day time, high average biomass productivities of 28-31 g m-2 d-1 and photosynthetic efficiencies of 3.5-4.3% were achieved in the continuous dilution experiments. In the repeated batch experiments, microalgal cells likely experienced stress due to sudden changes in light and temperature after daily dilution. Oxygen production measurements were performed at different temperatures to determine the maximum photosynthetic rates of the reactor cultures. Dense cultures of Picochlorum sp. (BPE23) were characterized by high maximum photosynthetic rates compared to previously reported data for Picochlorum and rates of other microalgae, particularly at a temperature of 39\u00b0C. In other oxygen evolution experiments and lab-based growth experiments in flat panel photobioreactors, Picochlorum sp. (BPE23) was also found to be characterized by a high specific growth rate and a low maintenance rate compared to other microalgal species, such as Neochloris oleoabundans and Nannochloropsis sp. (Chapter 3). The results in Chapter 3 and 5 highlight the robustness and potential of Picochlorum sp. (BPE23) for fast growth under changing outdoor conditions.\n\nIn Chapter 5, the possibility of passive temperature control in a novel V-shaped photobioreactor is investigated for a low-latitude location. A mathematical temperature model was developed to simulate the reactor temperature and relevant heat fluxes during the year on Bonaire. Two scenarios were simulated; one in which the V-shaped photobioreactor is used on land, and one in which the photobioreactor floats on water in a basin. The average temperature of the land-based photobioreactor was found to be 35\u00b0C, while the average temperature of the floating photobioreactor was found to be 32\u00b0C. Nevertheless, the temperature fluctuation in a floating photobioreactor (i.e. 30-33\u00b0C) was found to be much smaller than that in the land-based reactor (i.e. 30-42\u00b0C) due to the buffering effect of the water body. In a worst-case scenario, in which convection above and below the reactor is assumed to be limited, the temperature in a floating photobioreactor was found to remain largely below 40\u00b0C, whereas the temperature in the land-based reactor was found to fluctuate up to 48\u00b0C. Still, for both the land-based and floating photobioreactors, the estimated reactor temperatures were found to be suitable for the cultivation of mesophilic and temperature-tolerant microalgae such as Picochlorum sp. without temperature control.\n\nIn Chapter 6, the key findings of this thesis are presented and discussed. Additionally, the maximum theoretical biomass productivity of Picochlorum sp., Neochloris oleoabundans, and Nannochloropsis sp. in a large-scale V-shaped photobioreactor located on Bonaire are estimated. This estimation was based on the biological parameters determined in Chapter 3 and the mathematical model developed in Chapter 2. These productivities were found to be 46.5, 49, and 39 g m-2 d-1, respectively, demonstrating the high growth potential of the Bonarian Picochlorum strain. The results highlight the importance of knowing the light intensity and temperature to which microalgal cultures are exposed in a specific cultivation system and location during the day and year. Based on this knowledge, microalgal strains should be selected that perform optimally under those conditions to maximize biomass productivity.\n\nIn a final step, the novel V-shaped photobioreactor is evaluated in Chapter 6 by means of a techno-economic model and SWOT analysis. The biomass production costs for Picochlorum sp. (BPE23) in a V-shaped photobioreactor on Bonaire were estimated to be 2.24 \u20ac kg-1 of dry weight at a production scale of 100 ha and 7.05 \u20ac kg-1 of dry weight at a scale of 1 ha. These costs were lower than the biomass production costs in other systems at the same location, due to the relatively high photosynthetic efficiency that can be achieved in a V-shaped photobioreactor and the absence of active temperature control. To lower the biomass production costs even further and to advance the commercialization of microalgal biomass, several opportunities must be investigated further such as the use of renewable energy (e.g. solar and\/or wind energy), the use of direct air capture or flue gas as a source of CO2, and the development of biorefinery approaches and higher added-value products or experiences.
