Bij zoogdieren kunnen de twee dominante ruggengraten, Neu5Ac en Neu5Gc, O-geacetyleerd worden op C4, C7, C8 en\/of C9, waardoor een familie positionele isomeren ontstaat van mono- tot tetra-O-acetyl. Deze modificaties sturen receptorinteractie, gevoeligheid voor sialidasen en virale hechting, en vormen zo de immuunherkenning, gastheer-microbe-interacties en tumorbiologie. Toch zijn deze processen op moleculair niveau moeilijk te bestuderen door de chemische labiliteit van de O-acetylesters, met name migratie tussen C7 en C9 en hydrolyse, en door de schaarste aan goed gedefinieerd materiaal uit natuurlijke bronnen. Dit proefschrift beschrijft synthetische benaderingen voor complexe sialoglycanen met gedefinieerde O-acetyleringspatronen en beoordeelt hun eigenschappen in biologisch relevante contexten.\n\nHoofdstuk 1 vat de structurele diversiteit van O-geacetyleerde Sia\u2019s samen over de Neu5Ac- en Neu5Gc-ruggengraten met substitutie op C4, C7, C8 en C9; het brengt hun spreiding in kaart over weefsels, soorten en ontwikkelingsstadia; en schetst de biosynthetische en \u201cediting\u201d-machinerie (CASD1, SIAE en virale hemagglutinine-esterasen), samen met de functionele consequenties voor receptorinteractie, sialidasengevoeligheid, viraal tropisme, immuunregulatie en tumorbiologie. Het biedt ook een beknopt overzicht van chemische en chemo-enzymatische routes naar siaalzuren en complexe sialoglycanen, met nadruk op beschermgroepsturing, late-stage acetyl-\u201cediting\u201d en modulaire enzymatische assemblage die positie-gedefinieerde O-geacetyleerde varianten opleveren; tevens wordt uitgelegd hoe esterlabiliteit, de migratie van C7 naar C9 en de beperkte hoeveelheid natuurlijk materiaal hebben het experimentele ontwerp vormgegeven. Ondanks deze vooruitgang ontbreken nog breed toepasbare en robuuste methoden voor de bereiding van architectonisch complexe O-geacetyleerde sialoglycanen.\n\nGD3 is een siaalzuur-bevattend ganglioside dat berucht lastig te verkrijgen is in goed gedefinieerde, O-geacetyleerde vormen vanwege lage natuurlijke abundanties, microheterogeniteit en de labiliteit van O-acetylesters. O-geacetyleerd GD3 is een prominent oncofoetaal ganglioside waarvan O-acetylering receptorherkenning, sialidasengevoeligheid en apoptosesignalering moduleert en dat in verband is gebracht met tumorprogressie, virale hechting en immuunregulatie. Hoofdstuk 2 beschrijft een eenvoudige chemo-enzymatische strategie die beperkte beschikbaarheid omzeilt en een beknopte synthese levert van 7-O-, 9-O- en 7,9-di-O-geacetyleerde GD3-gangliosiden. De aanpak omvat chemische assemblage van een gemeenschappelijke tetrasaccharide-precursor als \u03b1-glycosylfluoride, koppeling aan sfingosine via een glycosynthase (EGCII E351S\/D314Y) en daaropvolgende O-acetyl-\u201cediting\u201d met coronavirale hemagglutinine-esterasen (BCoV en MHV-S). Deze route omzeilt sterk-basische stappen uit eerdere GD3-syntheses die onverenigbaar zijn met acetylesters. Elk acetyleringspatroon wordt volledig gekarakteriseerd en vervolgens gebruikt om het oppervlak van humane erytrocyten te herconfigureren. LC-IM-MS bevestigde presentatie in het membraan met behoud van het acetyl-esterpatroon, inclusief het base-gevoelige 7-O-geacetyleerde GD3. De gemodificeerde erytrocyten zijn daarna ge\u00ebvalueerd in label-vrije hemagglutinatie-assays met nanodeeltjes die de HKU1-spike presenteren, wat patroon-specifieke binding onder membraan-achtige omstandigheden liet zien, consistent met preferenti\u00eble herkenning van 9-O-geacetyleerd siaalzuur. Deze vooruitgang overwint cruciale synthetische en presentatie-barri\u00e8res en opent een route naar systematische, membraancontext-studies van O-acetylering-afhankelijke herkenning bij infectie en immuniteit.\n\nPolysiaalzuur (PSA), bijvoorbeeld in de capsule van Escherichia coli K1, is vaak O-geacetyleerd. Mechanistische analyse is echter beperkt door het ontbreken van plaats-gedefinieerde standaarden die 7-O- van 9-O-acetylering over de keten onderscheiden. Hoofdstuk 3 ontwikkelt een chemo-enzymatische strategie voor de assemblage van E. coli K1-antigeenfragmenten met drie of vier O-geacetyleerde siaalzuureenheden. De route gebruikt lineaire chemische opbouw van meervoudig O-geacetyleerde tri- en tetrasialosiden, gevolgd door enzymatische O-acetyl-\u201cediting\u201d met coronavirale hemagglutinine-esterasen (BCoV en MHV-S) om varianten te genereren met 7-O-acetyl of 9-O-acetyl op alle Sia-residuen. Met deze positie-gedefinieerde probes toont ELISA aan dat humaan L-ficolin 7-O-geacetyleerde boven 9-O-geacetyleerde en niet-gemodificeerde oligosiaalzuren preferentieel herkent. STD-NMR en moleculaire modellering wijzen erop dat de methylgroep van sialaat-7-O-acetyl uitlijnt met hydrofobe residuen in L-ficolin, wat een extra hydrofoob contact oplevert bovenop de interacties van het siaalzuur-skelet. Deze resultaten leggen positie-specifieke herkenning van O-geacetyleerd PSA in de aangeboren immuniteit vast en bieden hulpmiddelen voor systematische studie van O-acetylering in gastheer-pathogeeninteracties.\n\nHoofdstuk 4 richt zich op het enzymatische apparaat dat verantwoordelijk is voor de installatie van N-glycanen, met bijzondere nadruk op oligosaccharyltransferase (OST), het membraaneiwit dat lipide-gekoppelde oligosacchariden (LLOs) in \u00e9\u00e9n stap overbrengt op asparagine-residuen binnen Asn-X-Ser\/Thr-motieven. Het mechanistisch begrip van OST en aanverwante stappen, inclusief substraatherkenning door de RFT1-flippase, is beperkt gebleven door de ontoegankelijkheid van native LLOs. Om dit te overwinen, is een chemo-enzymatische benadering ontwikkeld om zowel native als enzymatisch gestabiliseerde pyrofosforothioaat-analogen te synthetiseren. De Man\u2083GlcNAc\u2082-kern werd verkregen uit sialoglycopeptide (SGP) via opeenvolgende glycosidase-trimming, omgezet in fosfaat- of fosforothioaat-donoren en gekoppeld aan chemisch gesynthetiseerde dolicholfosfaat. Het platform biedt een basis voor mechanistisch inzicht en kan de ontwikkeling van selectieve OST-modulatoren voor therapeutische toepassingen ondersteunen.","summary":"O-acetylated sialic acids are derivatives of the nine-carbon keto-sugar sialic acid that commonly terminate vertebrate glycans. In mammals, the two predominant backbones, Neu5Ac and Neu5Gc, can be O-acetylated at C4, C7, C8 and\/or C9, generating a family of positional isomers from mono- to tetra-O-acetyl. These modifications tune receptor engagement, sialidase susceptibility, and viral attachment, thereby shaping immune recognition, host-microbe interactions, and tumor biology. Here, processes have been difficult to study at a molecular level due to the chemical lability of the O-acetyl esters, especially migration among C7 to C9 and hydrolysis, and by the scarcity of well-defined material from natural sources. This thesis describes synthetic approaches to complex sialoglycans bearing defined O-acetylation patterns and assesses their properties in biologically relevant contexts.\n\nChapter 1 summarizes the structural diversity of O-acetylated Sias across the Neu5Ac and Neu5Gc backbones with substitution at C4, C7, C8, and C9; maps their distribution across tissues, species, and developmental stages; and outlines the biosynthetic and editing machinery (CASD1, SIAE, and viral hemagglutinin esterases) together with functional consequences for receptor engagement, sialidase susceptibility, viral tropism, immune regulation, and tumor biology. It also provides a concise overview of chemical and chemoenzymatic routes to sialic acids and complex sialoglycans, highlighting protecting group control, late stage acetyl editing, and modular enzymatic assembly that deliver position defined O-acetylated variants, and it explains how ester lability, C7 to C9 migration, and limited natural material have shaped experimental design. Despite these advances, broadly applicable and reliable methods for preparing architecturally complex O-acetylated sialoglycans remain insufficient.\n\nGD3 is a sialic acid-containing ganglioside that is notoriously difficult to access in well-defined, O-acetylated forms because of low natural abundance, microheterogeneity, and the lability of O-acetyl esters. O-acetylated GD3 is a prominent oncofetal ganglioside whose O-acetylation modulates receptor recognition, sialidase susceptibility, and apoptosis signaling, and has been implicated in tumor progression, viral attachment, and immune regulation. In Chapter 2, a facile chemoenzymatic strategy was devised to overcome limited access and provide concise synthesis of 7-O-, 9-O-, and 7,9-di-O-acetylated GD3 gangliosides. The approach comprises chemical assembly of a common tetrasaccharide precursor as an \u03b1-glycosyl fluoride, coupling to sphingosine by a glycosynthase (EGCII E351S\/D314Y), and subsequent O-acetyl editing with coronaviral hemagglutinin-esterases (BCoV and MHV-S). This route circumvents strong-base steps in earlier GD3 synthesis that are incompatible with acetyl esters. Each acetylation pattern is fully characterized and then used to remodel the surface of human erythrocytes. LC-IM-MS confirmed membrane presentation with preservation of the acetyl-ester pattern, including the base-sensitive 7-O-acetylated GD3. The engineered erythrocytes were subsequently evaluated in label-free hemagglutination assays using nanoparticles displaying the HKU1 spike, revealing pattern-specific binding under membrane-like conditions consistent with preferential recognition of 9-O-acetylated sialic acid. These advances overcome key synthetic and presentation barriers and open a route to systematic, membrane-context studies of O-acetylation dependent recognition in infection and immunity.\n\nPolysialic acid (PSA), exemplified by the Escherichia coli K1 capsule, is frequently O-acetylated. However, mechanistic analysis has been limited by the absence of site-defined standards distinguishing 7-O- from 9-O-acetylation across the chain. Chapter 3 develops chemoenzymatic strategy for assembly of Escherichia coli K1 antigen fragments containing three or four O-acetylated sialic acid units. The route uses linear chemical construction of multiply O-acetylated tri- and tetrasialosides, followed by enzymatic O-acetyl editing with coronaviral hemagglutinin-esterases (BCoV and MHV-S) to generate variants bearing 7-O-acetyl or 9-O-acetyl across all Sia residues. With these position-defined probes, ELISA demonstrates preferential recognition of 7-O-acetylated over 9-O-acetylated and non-modified oligosialic acids by human L-ficolin. STD-NMR and molecular modeling indicate the methyl of sialate-7-O-acetyl aligns with hydrophobic residues in L-ficolin, providing an additional hydrophobic contact beyond interactions made by the sialic-acid scaffold. These results establish position-specific recognition of O-acetylated PSA in innate immunity and provide tools for systematic study of O-acetylation in host-pathogen interactions.\n\nChapter 4 focuses on the enzymatic machinery responsible for N-glycan installation, with particular emphasis on oligosaccharyltransferase (OST), the membrane enzyme that transfers lipid-linked oligosaccharides (LLOs) en bloc to asparagine residues within Asn-X-Ser\/Thr motifs. Mechanistic understanding of OST and related steps, including substrate recognition by the RFT1 flippase, has been limited by the inaccessibility of native LLOs. To overcome this, a chemoenzymatic approach was developed to synthesize both native and enzymatically stabilized pyrophosphorothioate analogs. The Man\u2083GlcNAc\u2082 core was obtained from sialoglycopeptide (SGP) via sequential glycosidase trimming, converted into phosphate or phosphorothioate donors, and coupled to chemically synthesized dolichol phosphate. This workflow provides well-defined, stable LLO substrates in quantities suitable for biochemical and structural studies, enabling detailed interrogation of OST substrate specificity and early steps in N-glycosylation.
