Dit proefschrift pakt deze beperking aan door middel van nanoschaal chemische en structurele karakterisering van polymeren en functionele oppervlakken. Door atoomkrachtmicroscopie (AFM) te combineren met akoestisch-mechanisch onderdrukte fotothermische infrarood nanospectroscopie (AMS-AFM-IR), ontrafelt dit proefschrift lokale heterogeniteit, hi\u00ebrarchische structurele organisatie en intermoleculaire interacties voorbij bulk-gemiddelde metingen.\n\nHoofdstuk 2 stelt een op AFM gebaseerde methodologie vast voor de schatting van het molecuulgewicht van individuele oppervlak-ondersteunde polymeerelementen op basis van kenmerken. Individuele polymeerkenmerken worden geanalyseerd via hun schijnbare oppervlakte en hoogte, gevolgd door een op dichtheid gebaseerde conversie naar molecuulgewicht. De resultaten laten zien dat oppervlak-ondersteunde polymeersystemen bestaan uit discrete kenmerken die verschillen in morfologie, molecuulgewicht en aggregatietoestand, en daarom niet kunnen worden behandeld als equivalente eenheden binnen \u00e9\u00e9n gemiddelde populatie. Op AFM gebaseerde analyse behoudt molecuulgewichtsdistributies op het niveau van individuele kenmerken voorbij bulk-gemiddelde metingen en identificeert geclusterde populaties die consistent zijn met verschillende aggregatietoestanden. Hoofdstuk 2 laat daarom zien dat variatie op nanoschaal een intrinsiek kenmerk is van het polymeermonster in plaats van een afwijking van de gemiddelde beschrijving.\n\nHoofdstuk 3 laat zien dat chiraliteit in chirale en racemische polymeersystemen ontstaat over moleculaire, enkelvoudige keten- en supramoleculaire lengteschalen. Bulk Chiraal-HPLC, circulair dichro\u00efsme (CD), infrarood (IR) en UV-Vis spectroscopie bieden gemiddelde informatie, maar AFM-beeldvorming gecombineerd met AMS-AFM-IR lost de nanoschaal chemisch-structurele heterogeniteit van deze systemen direct op. Chiraliteit wordt niet uitgedrukt als een enkele uniforme gemiddelde toestand, maar als de som van lokaal verschillende conformaties, assemblagetoestanden en chemische signaturen op nanoschaal. Centrale chiraliteit is aanwezig op moleculair niveau, terwijl polymerisatie aanleiding geeft tot ruggengraatchiraliteit in het chirale polymeer en supramoleculaire chiraliteit in het racemische polymeer. Hoofdstuk 3 toont daarom aan dat chiraliteit hi\u00ebrarchisch kan worden opgelost over verschillende organisatieniveaus.\n\nHoofdstuk 4 onderzoekt zelf-geassembleerde monolagen gevormd door monomeren en polymeren op hooggeordend pyrolytisch grafiet (HOPG). AFM en AFM-IR laten zien dat het monomeersysteem een bedekkings-invariante monolaag vormt, terwijl het polymeersysteem een co\u00f6peratieve monolaag vormt met bedekkingsafhankelijke structurele veranderingen, laterale associatie en een opkomende carbonylband bij ongeveer 1725 cm\u207b\u00b9. Samen met de toename in watercontacthoek leveren deze resultaten direct bewijs dat de polymeermonolaag wordt beheerst door adsorbaat-adsorbaat intermoleculaire interacties, terwijl de monomeermonolaag gedomineerd blijft door substraat-adsorbaat interacties. Hoofdstuk 4 laat daarom zien dat lokale monolaagorganisatie en intermoleculaire interacties de vorming van deze functionele oppervlakken sturen.\n\nSamenvattend laat dit proefschrift zien dat polymeren en functionele oppervlakken het best begrepen kunnen worden voorbij bulk-gemiddelde karakteriseringsmethoden en op het niveau van lokale organisatie. Over individuele polymeerkenmerken, supramoleculaire assemblages en zelf-geassembleerde monolagen wordt aangetoond dat lokale heterogeniteit, hi\u00ebrarchische structurele organisatie en intermoleculaire interacties bepalen hoe organisatie en eigenschappen ontstaan. Over het geheel genomen vestigt dit proefschrift nanoschaal chemisch-structurele karakterisering als een directe route naar het begrijpen van polymeren en functionele oppervlakken op het niveau waar hun relevante organisatie en eigenschappen worden gevormd.","summary":"Polymers and functional surfaces are often described by bulk-average characterization methods, but these do not fully capture how organization and properties arise across hierarchical length scales. This thesis addresses this limitation through nanoscale chemical and structural characterization of polymers and functional surfaces. By combining atomic force microscopy (AFM) with acoustic-mechanical suppressed photothermal infrared nanospectroscopy (AMS-AFM-IR), this thesis resolves local heterogeneity, hierarchical structural organization, and intermolecular interactions beyond bulk-average readouts.\n\nChapter 2 establishes an AFM-based methodology for single-feature-resolved molecular-weight estimation of surface-supported polymer features. Individual polymer features are analyzed through their apparent area and height, followed by density-based conversion to molecular weight. The results show that surface supported polymer systems are composed of discrete features that differ in morphology, molecular weight, and aggregation state, and therefore cannot be treated as equivalent units within one averaged population. AFM-based analysis preserves feature-level molecular-weight distributions beyond bulk-averaged readouts and resolves clustered populations consistent with different aggregate states. Chapter 2 therefore shows that nanoscale variation is an intrinsic feature of the polymer sample rather than a deviation from its average description.\n\nChapter 3 shows that chirality in chiral and racemic polymer systems emerges across molecular, single-chain, and supramolecular length scales. Bulk Chiral-HPLC, circular dichroism (CD), infrared (IR), and UV-Vis spectroscopy provide average information, but AFM imaging combined with AMS-AFM-IR resolves the nanoscale chemical-structural heterogeneity of these systems directly. Chirality is not expressed as a single uniform average state, but as the sum of locally distinct conformations, assembly states, and nanoscale chemical signatures. Central chirality is present at the molecular level, while polymerization gives rise to backbone chirality in the chiral polymer and supramolecular chirality in the racemic polymer. Chapter 3 therefore demonstrates that chirality can be resolved hierarchically across different levels of organization.\n\nChapter 4 investigates self-assembled monolayers formed by monomers and polymers on highly ordered pyrolytic graphite (HOPG). AFM and AFM-IR show that the monomer system forms a coverage-invariant monolayer, whereas the polymer system forms a cooperative monolayer with coverage-dependent structural changes, lateral association, and an emergent carbonyl band at approximately 1725 cm\u207b\u00b9. Together with the increase in water contact angle, these results provide direct evidence that the polymer monolayer is governed by adsorbate-adsorbate intermolecular interactions, whereas the monomer monolayer remains dominated by substrate-adsorbate interactions. Chapter 4 therefore shows that local monolayer organization and intermolecular interactions govern the formation of these functional surfaces.\n\nTaken together, this thesis shows that polymers and functional surfaces are best understood beyond bulk-average characterization methods and at the level of local organization. Across single polymer features, supramolecular assemblies, and self-assembled monolayers, local heterogeneity, hierarchical structural organization, and intermolecular interactions are shown to govern how organization and properties arise.
